Pharmacological characterization of antineoplastic alkylphosphocholines

چکیده
The Unit of Toxicology and Chemotherapy originated in 1993 from the former Division of Carcinogenesis and Chemotherapy. The objective of this unit is to better understand mechanisms of toxicity and carcinogenesis of drugs and chemicals and to develop improved methods for diagnosis and treatment of cancer. In this context the mechanism of action as well as the spectrum of anticancer and anti-protozoan activities of alkylphosphocholines was studied. In addition, colorectal micro-metastases as well as disseminated tumor cells (DTC) were identified by using sensitive assays. These included a PCR-RFLP assay for K-ras mutations in DTC that were shed from K-ras mutated colon carcinoma, as well as RT-PCR assays for cytokeratin 20 and guanylylcyclase C. Finally, models of liver metastasis of colorectal cancer as well as of bone metastasis of breast cancer were developed and used for evaluating new therapies. Separation and quantification of alkylphosphocholines by reversed phase high performance liquid chromatography.
Alkylphosphocholines represent a new class of drugs with remarkable antineoplastic and antiprotozoal activity. For instance, hexadecylphosphocholine has been approved for the topical treatment of skin metastasis. In addition, it was successfully studied in India for the treatment of leishmaniasis. Different phase-I and phase-II-trials resulted in cure rates of more than 97%. To optimize antitum...

Interdisciplinary cooperation between basic and clinical research has resulted in the discovery and development of alkylphosphocholines, a new class of substances for the treatment of breast cancer. In contrast to most antitumor substances, the alkylphosphocholines do not attack the cell nucleus, but the cell membrane. This report presents a systematic study which, for the first time, provides ...

Free-living amoebae of the genus Acanthamoeba are causing serious chronic conditions such as destructive keratitis in contact lens wearers or granulomatous amoebic encephalitis in individuals with compromised immune systems. Both are characterized by the lack of availability of sufficiently effective and uncomplicated, manageable treatments. Hexadecylphosphocholine (miltefosine) is licensed for...
متن کاملReduced Expression of the Retinoblastoma Protein Shows That the Related Signaling Pathway Is Essential for Mediating the Antineoplastic Activity of Erufosine
Erufosine is a new antineoplastic agent of the group of alkylphosphocholines, which interferes with signal transduction and induces apoptosis in various leukemic and tumor cell lines. The present study was designed to examine for the first time the mechanism of resistance to erufosine in malignant cells with permanently reduced expression of the retinoblastoma (Rb) protein. Bearing in mind the ...
متن کاملBiological Monitoring of Healthcare Workers Exposed to Antineoplastic Drugs: Urinary Assessment of Cyclophosphamide and Ifosfamide
Exposure of health care workers to antineoplastic drugs and subsequent adverse health effects is still an open issue. Very little has been studied on the extent of occupational exposure and handling conditions of antineoplastic drugs in Iran. We aimed to determine cyclophosphamide and ifosfamide concentrations in the urine samples of oncology healthcare workers. In addition, we assessed workpla...
